Program P r o j e c t s on t h e Biology o f t h e Immune System r e p r e s e n t a n award mechanism which t h e Branch h a s employed t o meet t h i s o b j e c t i v e . RESEARCH GOALS AND SCOPE The g o a l o f t h e s e Program P r o j e c t s i s t h e a t t a i n m e n t of a complete u n d e r s t a n d i n g o f t h e s t r u c t u r e and f u n c t i o n o f t h e immune system and i t s p r o d u c t s , i t s i n t e r a c t i o n w i t h o t h e r body s y s t e m s , and f u l l knowledge o f t h e g e n e t i c and o t h e r f a c t o r s which r e g u l a t e i t s development and f u n c t i o n . An u l t i m a t e p r a c t i c a l a p p l i c a t i o n of t h i s information is t h e use of s e l e c t e d cloned c e l l s o f t h e system, o r t h e i r products, f o r t h e c l i n i c a l c a r e o f r e c o n s t i t u t i o n o f immunodeficient i n d i v i d u a l s , t o a l l e v i a t e a l l e r g i c s t a t e s , t o p r o v i d e r e s i s t a n c e t o l i f e - t h r e a t e n i n g i n f e c t i o n s and t o c o r r e c t a b e r r a n t o r d e f e c t i v e immunoregulatory mechanisms. The scope o f t h e s e program p r o j e c t s i n c l u d e s s t u d i e s o f e v e r y f a c e t o f t h e immune r e s p o n s e , r a n g i n g from t h e i n i t i a l s t e p o f a n t i g e n r e c o g n i t i o n t o t h e f i n a l e l a b o r a t i o n o f immunologically d i s t i n c t i v e p r o d u c t s o f s p e c i f i c immunocytes. Research c u r r e n t l y s u p p o r t e d by t h i s mechanism was d e s i g n e d t o expand knowledge o f t h e morphologic and f u n c t i o n a l h e t e r o g e n e i t y o f lymphocyte p o p u l a t i o n s and d e v e l o p t h e c a p a b i l i t y f o r i d e n t i f i c a t i o n and s e l e c t i o n of lymphocyte s u b p o p u l a t i o n s , w i t h s p e c i f i c immune r e a c t i v i t y o r m o l e c u l a r c o m p o s i t i o n , f o r u s e i n s o m a t i c h y b r i d i z a t i o n o f such p o p u l a t i o n s and s e l e c t i v e p r o d u c t i o n o f s p e c i f i c , b i o l o g i c a l l y a c t i v e , lymphocyte p r o d u c t s . S i m i l a r s t u d i e s o f macrophages, o t h e r a c c e s s o r y and e f f e c t o r c e l l s , and networks o f c e l l s and m o l e c u l e s t h a t a f f e c t t h e a c t i v a t i o n , d i f f e r e n t i a t i o n and r e g u l a t i o n o f c e l l s o f t h e immune system are a p p r o p r i a t e . OBJECTIVES AND SCOPE The o b j e c t i v e o f t h e DRTCs i s t o b r i n g t o g e t h e r i n v e s t i g a t o r s from r e l e v a n t d i s c i p l i n e s i n a manner which w i l l enhance and e x t e n d t h e e f f e c t i v e n e s s o f r e s e a r c h and t r a i n i n g b e i n g conducted i n t h e f i e l d o f d i a b e t e s and i t s c o m p l i c a t i o n s . A D i a b e t e s C e n t e r must be an i d e n t i f i a b l e u n i t w i t h i n a s i n g l e u n i v e r s i t y medical c e n t e r or a c o n s o r t i u m o f c o o p e r a t i n g i n s t i t u t i o n s , i n c l u d i n g an a f f i l i a t e d u n i v e r s i t y . When f u l l y developed, t h e DRTCs are e x p e c t e d t o encompass t h e f o l l o w i n g : 1 ) f a c i l i t a t i n g and s t r e n g t h e n i n g b a s i c and c l i n i c a l r e s e a r c h r e l a t e d t o d i a b e t e s and i t s c o m p l i c a t i o n s ; 2 ) t r a i n i n g p o s t d o c t o r a l f e l l o w s t o conduct d i a b e t e s - r e l a t e d r e s e a r c h ; 3) t r a i n i n g h e a l t h p r o f e s s i o n a l s a b o u t d i a b e t e s and i t s management; 4 ) d e v e l o p i n g a model d e m o n s t r a t i o n f a c i l i t y t o c o n t r i b u t e t o t h e above endeavors; and, 5 ) t r a n s f e r r i n g advances i n t h e f i e l d o f d i a b e t e s i n t o improved care f o r p e o p l e w i t h d i a b e t e s . A l l o f t h e s e areas need n o t be developed t o t h e same d e g r e e . However, a s t r o n g b a s e o f b i o m e d i c a l r e s e a r c h i s t h e most i m p o r t a n t f u n c t i o n o f a c e n t e r . Accordingly, a program of e x c e l l e n c e i n b i o m e d i c a l r e s e a r c h i n t h e area o f d i a b e t e s and r e l a t e d m e t a b o l i c and e n d o c r i n e d i s o r d e r s i n t h e form o f NIH-funded r e s e a r c h p r o j e c t s , program p r o j e c t s , or o t h e r peer-reviewed r e s e a r c h must be i n e x i s t e n c e a t t h e t i m e o f submission o f a C e n t e r a p p l i c a t i o n .
