									Probe Synthesis and Hybridization Request
Read the directions for the probe synthesis method you are requesting & fill in the sample
name column on page two. Send the saved document to microarray@mednet.ucla.edu
with the subject heading “Your Name, Service Request”


Submit Samples for Fluorescent Linear Amplification and Hybridization
Note: this service is for synthesis of cRNA probe, using Agilent’s Low RNA Input Fluorescent Linear
Amplification Kit, and hybridization of that probe to Agilent’s animal oligo arrays of 1array/slide format
only. If you would like to synthesize cDNA using this method or would like to hybridize probe to any
other array type or format then e-mail microarray@mednet.ucla.edu to make the proper arrangements prior
to filling out this sheet.

Sample Preparation
   Send or bring total RNA samples, on dry ice, to the core at a concentration of
500ng/ul and at a volume of at least 6ul. We start synthesis with 500ng of total RNA with
typical cRNA yeilds of 6 to 8ug depending on the tissue type and quality of starting total
RNA.


Submit Samples for Fluorescent Direct Labeling and Hybridization
Note: this service is for synthesis of cDNA probe, generated by Agielnt’s Fluorescent Direct Labeling
Kit, and hybridization of that probe to Agilent’s animal oligo arrays of 1array/slide format only. If you
would like to hybridize to a different array type or format e-mail microarray@mednet.ucla.edu to make the
proper arrangements prior to filling out this form.

Sample Preparation
    Send your total RNA samples, on dry ice, to the core at a concentration of 1ug/ul. We
start probe synthesis with 20ug total RNA and hybridize all labeled probe generated to
one array.


                           Directions for filling out service request table

A sample set indicates samples that will be hybridized together onto one array. Please
note the dye column. This column informs us of the dye you wish to be incorporated
into the probe generated from the corresponding total RNA sample. For each sample
we will assign a Hyb ID and a Sample ID then send the table back to you via e-mail.
Keep this table for your records and please label the top of your sample tubes
accordingly. Please also label the concentration on the side of each tube.

 If total RNA is to be used for more than one probe synthesis there is no need to aliquot
the same sample into different tubes. For example, if the assigned sample ID’s are 10018,
10019, 10020, 10021, then label the one sample tube as 100(18)(19)(20)(21). Keep in
mind that probe generation using the low input kit usually generates enough material for
several hybridizations where as each probe generated using the direct labeling method
must be used on one single array.
