Instructions for use
Intended use
PBS Buffer Concentrate is used as a diluent and for washing steps in immunohistochemical staining procedures.
It is intended for research use only.
Summary and explanations
Immunohistochemical staining procedures consist of sequential incubation steps with blocking solutions, antibodies and
secondary reagents, enzymes and chromogenic substrates carried out on tissue sections. Washing away the applied
reagents after each incubation step is essential for receiving optimal staining results. Zytomed Systems PBS Buffer
Concentrate is helpful for effective washing and good staining results.
Principle of the method
Zytomed Systems PBS Buffer Concentrate is a 33fold concentrated phosphate buffered saline. For preparation of the
working strength solution the buffer concentrate is diluted 1:33 with deionised or distilled water. The resulting solution
has a pH of 7.4.

Storage and handling
The solution should be stored at 2-8° Do not free ze it. Under these conditions the solution is stable up to the expiry
date indicated on the label. Do not use product after the expiry date.

reagent, please contact Zytomed Systems’ technical support or your local distributor.
PBS Buffer Concentrate is not considered hazardous material in terms of directive 67/548/EWG. No Material safety
data sheet and no marking according to the EG-directive are necessary.
Reagent preparation
Bring PBS Buffer Concentrate to room temperature prior to dilution.
Preparation of the PBS Buffer working strength solution:
   • Transfer the complete content of one 15 ml bottle PBS Buffer Concentrate into a 500 ml volumetric flask. If salt
       precipitates remain in the bottle, this must be dissolved and completely transferred to the volumetric flask too.
   • Fill up with deionised or distilled water to exactly 500 ml. Mix thoroughly.
   • The pH-value should be at 7.4.
15 ml PBS Buffer Concentrate + 485 ml distilled water = 0.5 L ready-to-use PBS-Puffer

Quality control
We recommend carrying out a positive and a negative control with every staining run. The positive control permits the
validation of appropriate processing of the sample. If the negative control has a positive result, this points to unspecific
staining. Please refer to the instructions of the detection system for guidance on general quality control procedures.
If you observe unusual staining or other deviations from the expected results please read these instructions carefully,
contact Zytomed Systems’ technical support or your local distributor. Also refer to the instructions of the detection
systems for guidance on general troubleshooting.
Limitations of the procedure
Immunohistochemistry is a complex technique involving both histological and immunological detection methods. It
requires a highly trained histotechnologist. Tissue processing and handling prior to immunostaining, for example
variations in fixation and embedding or the inherent nature of the tissue can cause inconsistent results (Nadji and
Morales, 1983). Inadequate counterstaining and mounting can influence the interpretation of the results.
If a detection system with alkaline phosphatase is used please note: larger amounts of PBS Buffer remaining on the
slides can lead to decreasing enzyme activity.

suitable for the intended use.
