SYSMEX CDA-500 FOR COUNTING AND SIZING OF CELLS AND PARTICLES THEORY Designed for ease of use The CDA-500 has been specifically designed for ease of use. By turning the power on, the CDA-500 automatically performs a self-check and cleaning cycle.To make a measurement simply put your sample in the measurement unit, recall a pre-defined measurement profile then press the COUNT key. The measurement is then completed in a matter of seconds. The CDA-500 can recall a range of pre-set measurement profiles. Different profiles can be set depending on the user's application, saving set-up time when making repeat sample measurements. System diagnostics – during operation the CDA-500 is constantly monitored for measurement errors or faulty operation. For example, if the aperture blocks, an error message informs the operator.. Measurement Principle The CDA-500 uses the electro-zone sensing method for sizing and counting particles.The detector assembly consists of an aperture (opening) through which suspended particles pass. Internal and external electrodes are on either side of the aperture.The detector assembly is immersed in an electrolyte that allows a constant current to flow between the electrodes. Non-conductive particles are suspended in an electrolyte.When a particle passes through the aperture during the measurement process, the particle decreases the current flow, increasing the resistance between the electrodes.The change in resistance causes a change in voltage between the electrodes proportional to the volume of the particle. The quantity of suspension drawn through the aperture is precisely controlled to allow the system to count and size the particles for an exact, reproducible volume. Several thousand particles can be counted and sized per second.This technique offers high sensitivity and excellent resolution because all particles are counted and sized.This method is independent of particle shape, colour, refractive index and density. Data Presentation The user interface provides an easy and convenient way of operating the instrument.The data analysis software enhances the flexibility of the instrument. The CDA-500 offers a variety of reports, from simple short form reports detailing the analysis conditions and key measured parameters, to full particle size distributions displayed in either graphic or tabular format. Analyzing results is simple with the range of data processing features available in the CDA-500 software. Measurements can be over-plotted, as either differential or cumulative data.A zoom facility enables the user to focus on a particular size range.The CDA-500 software allows the averaging of results and the automatic calculation of a variety of statistics. In addition a full range of variance graphs are available enabling the operator to quickly recognize any changes in particle size. The data can be easily exported for use in independent software packages. A key feature is the ability to integrate particle size data into reports by "cutting and pasting". Flexible The CDA-500 has a range of measurement modes that can be tailored to the analysis requirements. s Manometer Mode for measuring the number of particles per unit volume. Time Mode for taking measurements for a specified time. Recount Mode for automatically repeating measurements. s s s A Peak Count Mode for counting the number of particles to a pre-set limit. APPLICATIONS Applications The CDA-500 can be used in a variety of counting and sizing applications, such as tissue culture cells, blood cells, algae, yeast, plankton, suspensions and spermatozoa. Different aperture diameters are available, to enable the user to select the most suitable one for the size of particles to be analysed.
