Quantitative PCR REAL TIME PCR SYSTEM Real Time PCR

Quantitative PCR REAL-TIME PCR SYSTEM Real-Time PCR System The first personal high-performance, low-cost real-time PCR system With the Mx3000P™ system, we bring real-time PCR instrumentation to the personal market by offering a single system that provides high performance and full-feature design at a low cost. The instrument combines precision thermal control, a broad range optical system, and sophisticated software that is easy to learn and use. Most important, the Mx3000P system delivers outstanding results with all QPCR chemistries. Optimized for Use with Stratagene’s Brilliant® QPCR and QRT-PCR Reagents The Mx3000P system was designed for optimal performance with the use of our Brilliant® reagents. Brilliant QPCR and QRT-PCR reagents provide maximum versatility with high-quality, reproducible results. All of our reagent kits contain SureStart® Taq DNA polymerase, a hot-start version of Taq that minimizes amplification of non-specific PCR products. Whether you choose to optimize your reactions with a core reagent kit or prefer the convenience of a master mix, there is a Brilliant QPCR or QRT-PCR reagent kit for you. Advanced Optical System Design  Four channels with customizable filters  Extended excitation range supports all QPCR chemistries  Unparalleled multiplex results using optimized illumination selection Unsurpassed Real-Time PCR Results  Excellent uniformity and reproducibility  Superior sensitivity with seven orders of magnitude  Exceptional multiplex results with up to four targets Most Powerful Software Available  Real-time data monitoring and evaluation  Enhanced algorithms for precise data analysis  Melting curve protocols for accurate product identification Advanced Optical System for Ultimate Performance Excitation filter wheel Halogen lamp Emission filter wheel Coaxial fiber optic cable 1 blank filter position for calibration Read head scans over each well, filter wheels rotate between scans 96-well plate Unsurpassed Real-Time PCR Results Excellent Uniformity The thermal cycler in the Mx3000P system has outstanding thermal uniformity with only +/-0.25 ˚C variation in sample temperature across the 96-well plate. The temperature range of the thermal block is 25-95 ˚C. The thermal ramp rate is dynamically controlled by the software and can ramp up to 2.5 ˚C per second. Outstanding Uniformity SYBR® Green I uniformity run using a plasmid target and detection with target-specific primers and Stratagene’s Brilliant® SYBR Green Master Mix. Exceptional Multiplexing The Mx3000P system is specifically designed to ensure optimal results with multiplex assays using up to four targets in the same sample. The broad excitation range enables the use of dyes with maximal spectral separation. Emission from reporter dyes in the sample are collected by fiber optics, spectrally filtered, and detected by a photomultiplier tube (PMT). Narrow bandpass filters ensure finely delineated fluorescent signals and minimal noise. All of these components work together to provide extraordinary linear response for targets in singleplex and in multiplex. A Superior Sensitivity The Mx3000P system achieves linearity of amplification over seven orders of magnitude, demonstrating the quality of fluorescence detection in the instrument. The Mx3000P system offers impressive discrimination with the ability to detect two-fold differences in initial template quantity with 99.7% confidence. Enhanced Sensitivity B Serial dilution of human genomic DNA. 5x107 to 50 genomic equivalents in 10-fold steps (n=3). Detection of ß-Actin using specific primers, a Cy5-labeled Molecular Beacon, and Stratagene’s Brilliant® QPCR Master Mix. Unparalleled Multiplex Performance Standard curves of four-fold serial dilutions of cDNA prepared from Stratagene’s QPCR Human Reference Total RNA, performed in singleplex (Panel A) and in multiplex (Panel B). The targets amplified were eNOS (FAM), HFE (HEX), CFTR (Texas Red®), and Cyclophilin (Cy5) as the endogenous reference. Wide Linear Dynamic Range The Mx3000P™ system demonstrates excellent resolution over seven orders of magnitude. The regression of correlation (r2) was 1.00 and the slope of the curve was –3.39. Most Powerful Software Available Easy Setup and Flexibility to Make Changes The Mx3000P system software is so easy to use that you can learn the software and begin running experiments within minutes of installing your system. Plate setup and data analysis are fast and easy because your experiments are displayed in a 96-well format that matches the sample plate. The Quick Setup option saves you time by collecting data for all wells and all dyes—you can assign well information, reassign standards or change well destinations at your convenience—even while the experiment is running or after the run has finished. Real-Time Results and Data Analysis In addition to viewing the progress of your run in real time, you can also analyze your results as they are being generated. You have full access to raw data plots so you can assess PCR progress and elect to stop the experiment, saving valuable research time, or add cycles for further data collection and analysis. Many Choices for Data Display and Analysis You decide what data analysis format best suits your needs—amplification plots, scatter plots, melting curve profiles, fluorescent intensity values, and more. You can also decide how data will be analyzed: replicates may be treated individually or collectively, and data points for a given cycle can be used based on an average, minimum, maximum or last endpoint value. Application Specific Analysis Real-time allelic discrimination—calls by threshold cycle or by endpoint fluorescence values Molecular beacon melting curve—run and analyze melting curves with automatic calculation of melting temperature or recommended annealing temperature range Comparative quantitation—determine relative quantities of target sample with respect to a calibrator using corrections for PCR efficiency of the gene of interest and the normalizer gene SYBR ® Green I — verify homogeneity of PCR products using dissociation curves Fluorescence plate reads—use scatter plots, fluorescence intensity values or final call results to identify presence or absence of product Enhanced Algorithms for Standardized and Reproducible Results The Mx3000P system uses enhanced algorithms such as replicate averaging, moving average, adaptive baseline, and amplification-based threshold to provide the most reliable and consistent data. You will always find outstanding reproducibility since these analysis algorithms are unchanging and applied to each data set. Time-Saving Data Analysis You can view all dyes on a single amplification plot without the need to export data. You can also run multiple experiments on the same plate and analyze single wells or groups of wells independently of others. In addition, you can easily export any graph or data file directly to PowerPoint, Excel, image, or text files, enabling you to create presentations and reports quickly and easily.
