                            PROJECTS UNDERTAKEN

M.Sc Project
Title : Chemical Investigation on Curcuma zedoaria Rosc. And Curcuma
aeruginosa Roxb. from Malaysia.
        i)    Identification of essential oils from Curcuma zedoaria dan Curcuma
              aeruginosa.
        ii)   Extraction and isolation of natural compound from Curcuma zedoaria
              based on the result of bioassay test. Chemical Structure elucidation of
              isolated compound using Nuclear Magnetic Resonans (NMR)
              Spectroscopy, Infra Red Spectroscopy, UV Spectrophotometer and Mass
              Spectrometer. Identification of chemical components in Essential Oil of
              Curcuma zedoaria.
        iii)  Isolation of polar compound from Curcuma zedoaria using various
              absorbent of column chromatography (HP 20, Sephadex LH 20, ODS,
              Silica). Purifying polar compound from column chromatography using
              Recycle-preparative HPLC. All the techniques have been done at HEJ
              Research Ins. Of Chemistry, University of Karachi Pakistan, during
              the student exchange program.

ABSTRACT

A chemical studies was conducted on the fresh rhizomes of Curcuma aeruginosa and
Curcuma zedoaria and the dry rhizomes of C. zedoaria. The essential oils of the fresh
rhizomes of C. zedoaria (1.8%) and C. aeruginosa (1.8%) obtained by hydrodistillation,
was analysed by a combination of capillary GC and GC/MS. This resulted in the
identification of 12 and 23 components, comprising 74.7% and 69.7% of the rhizome oil
of C. zedoaria and C. aeruginosa respectively. Furanogermenone (45.2%) was found to
be the major constituent of C. zedoaria whilst curzerenone (33.2%) is the major
component for C. aeruginosa. The essential oil components were identified by
comparing their Kovats Indices with those reported in the literature and their mass
spectral data with those from the mass spectral database. An investigation on the
butanol and water extract of the dry rhizomes led to the isolation of four compounds
identified as zedoalactone B (1R, 4S, 5R, 10R)-1, 4, 10-trihydroxy-guaia-7(11), 8-dien-
12, 8-olide), three new compounds (1S, 4R, 5R, 10R)-4, 10-dihydroxyguai-7(11), 8-dien-
12, 8-olide and two epimers of zedoalactone A i.e (1S, 4R, 5R, 8R, 10S)-4,10-
dihydroxyguai-7(11)-en-12, 8-olide and (1S, 4S, 5R, 8R, 10S)-4, 10-dihydroxyguai-7(11)-
en-12, 8-olide. The above compounds were isolated through column chromatography
(using diaion HP 20 and sephadex LH 20 as adsorbents) and Recycle High Performance
Liquid Chromatography (RHPLC). Zedoalactone B was previously reported to exist in
the rhizomes of C. zedoaria. (1S, 4S, 5R, 10S)-4, 10-Dihydroxyguaia-7(11)8-dien-12, 8-
olide and the two epimers of zedoalactone A are newly reported compounds from C.
zedoaria which have not been reported before. The structures of isolated compounds
were identified through a detailed analysis of their nuclear magnetic resonance and
mass spectral data.
B.Sc Project
Title: Isolation of Active Component from the Crude Extract of Trema Orientalis
Stem Bark.
Isolation of the natural compound from the crude extract of local plant (Trema Orientalis)
using Chromatography Technique (Thin Layer Chromatography and Column
Chromatography). Interpretation of spectrometry data to determine the compound
structure (NMR, Mass Spect. IR Spectrometer).

Research Assistant at Interim Institute for Pharmaceutical and Nutraceutical
Bioteknology one of Biovalley Project funded by MOSTI. Responsible to incharge and
running two major equipments in the Interim Institute; Waters Micromass LCMS/MS Q-
TOF Micro and Recycled Preparative HPLC.

Waters Micromass LCMS/MS Q-TOF Micro is the first and the only one in Malaysia.
This equipment is one of the latest technology by Waters Micromass, combining
sensitivity and accuracy of molecular mass detection. The ability to produce the Exact
Mass of the compound give the very useful information in chemical compound structure
elucidation.

2003-2004
Part time tutor and Lab Demonstrator at School of Chemical Sciences and Food
Technology, Faculty of Science and Technology, UKM. Teaching Organic Chemistry and
Physical chemistry for first year students.

Research Attachment at H.E.J. Research Institute of Chemistry, University of
Karachi, Pakistan under supervision of Prof. Mohd. Iqbal Choudhary, Director (Acting)
of that institute for one and half month. Within that period, I have learned isolation
process for polar compound from buthanol extract and water extract. I have successfully
isolated 9 pure compound from both extracts using column chromatography technique
with HP20, Sephadex LH20 and silica as absorbent and Recycle-Preparative HPLC
machine.
