The TaqMan® Custom Array is a 384-well micro fluidic card (array) that enables you to perform 384 simultaneous real-time PCR reactions without the need for liquid-handling robots or multi-channel pipettors to load samples. This highly flexible array enables 1 to 8 samples to be run in parallel against 12 to 384 TaqMan® Gene Expression Assay targets (including manufacturing controls). The assays are pre-loaded into each of the array’s wells, easily facilitating your required throughput. Choose from over 50,000 inventoried TaqMan Gene Expression Assays designed for human, mouse, and rat genes to be loaded into a TaqMan Custom Array (Figure 1). The TaqMan®

Array is designed for use on the flexible Applied Biosystems 7900HT Fast RealTime PCR System fitted with a 7900HT TaqMan® Array Upgrade. Ultimate Microarray Validation Tool TaqMan Arrays are exactly the right tool for validating the tens or hundreds of hits that come from microarrays because they can be customized to include up to 384 of those hits in one easy-to-use array. Using individual assays, or even SYBR® dye-based assays, to look at 12, 48, or 96 targets can quickly become unmanageable and expensive. TaqMan Arrays enable researchers to accomplish the validation necessary to arrive at the right answer easily and affordably.

Ideal Screening Technology TaqMan Arrays are ideal for screening biomarkers and toxicology panels, and for analyzing pathways, target classes, and complete disease sets. Because TaqMan Arrays don’t require liquidhandling robots for loading, you get standardized results with low variability across multiple users and laboratories. Plus, TaqMan Gene Expression Assays— the benchmark of specificity and sensitivity in real-time PCR—are preloaded into the TaqMan Array, ensuring reliable performance. Create the Perfect Array You simply select TaqMan Gene Expression Assays and the optimal TaqMan Custom Array format for your experiment, and we will deliver your TaqMan Arrays pre-loaded with your selected assays. Choose 12 to 384 target assays from our collection of inventoried TaqMan Gene Expression Assays covering human, mouse, and rat genes. Ordering is easy with the online TaqMan Custom Array configuration tool to help you find and select genes and assays. TaqMan Custom Arrays are available in nine different formats covering 12, 16, 24, 32, 48, 64, 96 (two choices), and 384 assays. Designing a TaqMan Custom Array Customizing a TaqMan Array through the Applied Biosystems Web site (www.appliedbiosystems.com) is simple. Choose your array format and TaqMan Gene Expression Assays, select a quantity, and place your order—it’s as easy as that. Online selection of TaqMan Assays is simple: search by using common keywords like Entrez gene symbols, RefSeq accession numbers, or UniGene IDs (Figure 2a). You can narrow your search results using the Target Class filters relevant to your research area (Figure 2b). For example, if you want to find TaqMan Assays for caspase genes in the area of apoptosis, select the “Apoptosis” Target class filter and enter “caspase” in the Keyword Search box to return results for caspase genes (Figure 2c).

Alternatively, TaqMan® Gene Sets are “virtual” Gene Signature Arrays containing pre-defined lists of TaqMan Gene Expression Assays matching genes specific to a target class or pathway, and simplify configuration of a TaqMan Custom Array. Pre-designed Gene Signature Arrays Are Also Available If you don’t have time to compose a list of assays, use one of our pre-made TaqMan Arrays. We have created TaqMan® Gene Signature Arrays—predesigned, focused gene sets for many important target classes and pathways. Genes in each of the arrays have been culled from pathway analysis tools, published review papers, and collaborator and customer input. Select from a variety of TaqMan Gene Signature Arrays covering areas such as GPCRs, immune response, or protein kinases. You can also use our Endogenous Control Arrays to assess which housekeeping genes are best for your specific study. Simple and Straightforward Loading Once you receive your TaqMan Array, using it is fast and simple. The TaqMan Array has eight sample-loading ports, each connecting to 48 reaction wells. Simply combine 30–1,000 ng of cDNA sample with TaqMan® Universal PCR Master Mix (for each port), pipette sample mix into the TaqMan Array, centrifuge briefly to fill the wells, and seal the card. Within minutes, your 384-well TaqMan Array is ready to run on the Applied Biosystems 7900HT Fast Real-Time PCR System (Figure 3). Imagine the time and cost savings you will benefit from without having to set up individual reactions, not to mention the risk of introducing variability. Accurate, quantitative gene expression results are available when thermal cycling is complete. The reduced number of pipetting steps streamlines the reaction set-up process and eliminates the need for liquid-handling robotics. The Applied Biosystems 7900HT Fast Real-Time PCR System TaqMan Arrays are designed expressly for use with the Applied Biosystems 7900HT Fast Real-Time PCR System— a high-throughput, real-time PCR platform

that detects and quantitates nucleic acid sequences.
