INSTRUCTIONS FOR STORAGE, THAWING & INSEMINATION OF FROZEN SEMEN
The 5ml semen straws are transported in a ‘Dry-Shipper’ tank which keeps the internal
temperature at -196°C (or thereabouts) for 21 days from the date of shipment.
The Dry Shipper contains no pourable liquid nitrogen, as it is absorbed into its special lining.
Never raise the metal canister out of the tank unless transferring straws into a liquid nitrogen
bath or another tank as this will irreversibly damage the semen.
Generally, a cotton plug in the canister prevents damage to the straws during transit.
Equine semen is far more delicate and sensitive than bull semen
You will need a liquid nitrogen bath for handling the straws.
ALWAYS PRECOOL YOU TWEEZERS before grasping the straws.
Ensure the name on the straw corresponds with the stallion whom the mare is to be AI’d with.

Best conception rates are achieved when the mare is inseminated as close to the point of ovulation
Ideally the semen will be inseminated into the mare’s uterus within 1 minute of thawing.
Grasp the straw firmly & directly over the end of the sealing bead in the liquid nitrogen bath.
***Grasping over the straw itself will increase the possibility of one of the beads ‘blowing out,’ thus
loosing and contaminating the semen.***
Quickly transfer the straw into a 50°C water bath for 45 seconds, ensuring the straw is
Transfer straw immediately into a 37°C water bath for 15 seconds to 1 minute.
Dry straw thoroughly with a tissue as excess water is spermicidal.
Cut the sealing bead end of the straw with clean scissors.
Pour the 5ml thawed semen directly into a pre-warmed sterile syringe barrel.
Be sure to flush the AI pipette with 5ml of air to expel all of the semen during insemination.
Peak post thaw motility may be achieved anywhere from 30 seconds to 8 minutes, thus it is best
to assess motility after breeding the mare, using a fraction remaining in the AI straw (held in the
Please return all Dry-Shipper tanks as soon as possible using the supplied TNT consignment note
of which you will need to sign and date. Advice your local TNT agent who will arrange pick up
and delivery transport for you.
For any unused straws, please top up the tank with liquid nitrogen prior to transport.
