Enzymology of hemicellulose conversion. This post doc project is focusing on structuralfunctional investigations of enzymes (glycoside hydrolases) involved in plant biomass conversion. Internationally, much attention is currently dedicated on biotechnical hydrolysis of plant polysaccharides. In our group we have a focus on hemicelluloses, a group of substituted heteroglycans making up about 25% of plants. We are studying fundamental aspects of bmannanases, a-galactosidases, xylanases and their interaction with complex substrates. The project involves molecular biology, enzymology, biochemistry, structural biology and analytical 1 chemistry.

Post-Doc

The aim of the project is to clarify the enigmatic energy-coupling mechanism performed by respiratory Complex I, pivotal to cell energy metabolism. The postdoctoral fellow will measure transmembrane proton pumping by complex I subunits and the homologous mrp antiporter proteins under various conditions. To this end, spectroscopic equipment developed specifically for these studies, unique nanoprobes and genetic constructs are available. The postdoctoral fellow will work in a cross-disciplinary environment, with a project that spans molecular-biological methods and spectroscopy, and addresses a key 1 biochemical/biophysical problem.

Post-Doc

Viral genome is a long, charged molecule enclosed in a rigid protein shell of the capsid. This physical situation implies that the genome is highly stressed. The project involves a physical-chemical approach to understand as broadly as possible the state of stress of a long, semi-flexible, chain, whose length is hundreds of times larger than the dimension of the volume of the viral capsid. The success of the virus, i.e., the infectivity, depends on its genome being efficiently encapsidated in a process of virion synthesis. The goal of this project is to show that the size of the genome must be consistent with the size of the shell formed from self-assembly of the capsid protein and that it is the key to viral infectivity. We propose to do so by directly addressing 1. viral genome: its packaging and encapsidation, the kinds of pressures involved and 2. viral capsid: its packaging motor, capsid strength and selfassembly. The project also involves a study of the controlled self-assembly of viral 1 nanocontainers.
