					   17 Beta-Hydroxysteroid
Dehydrogenase in complex with
    estradiol and NADP+
    Presented by Bram Sebastian
               Introduction
• Short Chain Dehydrogenase family
• Catalyses reversible reduction of estrone to
  estradiol
• Play roles in the development of human
  breast cancer
• Precise orientation of substrate and cofactor
in the active site is important for inhibitor
  design and understanding catalytic process
                   History
• Structure determined in early 1990s by
  crystallography
• Enzyme extracted from the human placenta
• The gene consist of six exons and five introns
• Two major type, Type I (estrogenic) and Type II
  (androgenic)
• Both enzyme activity have been studied in
  other tissues (e.g. liver, prostate)
Enzyme Convert position 17 of steroid
        backbone structure
General Structure
Enzyme interact with estradiol
Active Site of Enzyme
Proposed reduction of estrone to
           esradiol
