FORMULATON AND IN VITRO PHARMACOLOGICAL SCREENING
OF TOPICAL GEL OF NABUMETONE AS AN NOVEL ANTI

Nabumetone is a Non-Steroidal Anti-inflammatory drug which has been extensively used in
relieving inflammation and analgesia. Nabumetone is non-acidic NSAID which is rapidly
metabolised in liver to a major active metabolite. The active metabolite of nabumetone inhibit
the enzyme preferentially, blocks cyclo-oxygnase-2 activity which is indirectly responsible
for the production of inflammation and pain during arthritis. The parent drug itself possess
anti inflammatory and analgesic activity. Presently nabumetone is available in the form of
tablet dosage form. But till date no topical gel formulation has been reported in the market.
The present research is an attempt to formulate and screen nabumetone in the gel form. Due
to high lipid solubility drug easily passes from stratum corneum and passes in to systemic
circulation; it avoids drug loss and produce anti-inflammatory activity. Further it is found
that, this drug have more bioavailability than any other NSAID. Of many drug delivery
systems the percutaneous drug delivery has been widely used as an advanced drug delivery
system and has some advantages in the controlled and sustained delivery of locally active
drugs. In order to obtain a sustained analgesic effect bioadhesive gels are used. In the present
study Carbopol 934 and HPMCK4M were used as a gel base, and their bioadhesive
properties were found to be beneficial. The prepared bioadhesive topical gel was evaluated
with the help of different parameters like drug content, spreadability, extrudability, swelling
index study, in–vitro drug diffusion study. Evaluation of nabumetone gel for topical anti-
inflammatory activity in Carrageenan induced rat hind paw edema model, demonstrated that
the nabumetone gel possess fairly good anti-inflammatory activity but not as good as
commercial product. Thus, nabumetone gel is a novel potential topical anti-inflammatory
