STUDIES ON INFLUENCE OF A NOVEL SOLUBLIZER IN LAMOTRIGINE
SUBLINGUAL TABLETS

The present work deals with the study of influence of a novel solublizer ‘SEPITRAP-
80TM’ in sublingual tablets of Lamotrigine. Lamotrigine is used in the treatment of
epilepsy. Lamotrigine is the model drug candidate with rationale to have rapid onset of
action in epilepsy. Sublingual route being highly vascularized with good flow of saliva is
effective in faster penetration and absorption of drugs serving noninvasive alternative to
injections. The aim was to have enhanced onset of action of Lamotrigine in the form of
sublingual tablets.
SEPITRAP-80TM was used as a solublizer in sublingual tablets in the concentration of 5,
10, 15, 20 and 25% of the tablet weight. Formulations were made with standard tablet
excipients and superdisintegrants. Superdisintegrant such as Sodium starch glycolate
were used in the concentration of 6% and 8%. Sublingual tablets were prepared by direct
compression method.
Formulations were evaluated for hardness, disintegration, in vitro drug release, in vivo
dissolution time, wetting time and water absorption test. Formulations using SEPITRAP-
80TM gives rapid release of drug. All the formulations showed satisfactory release within
5 minutes. Formulations with SEPITRAP-80TM gave t90% within 2 minutes.
SEPITRAP-80TM can be a promising solubilizer and an effective bioavailability enhancer
for poorly soluble drugs for various novel drug delivery systems.
