DNA Extraction Budget – As methylation (Glasauer)
4-Apr-11
Item Cost Supplier Description
Disposable tubes $150 Sigma Falcon tubes, PCR tubes
Aldrich
Antibiotics and medium supplements $100 Sigma Plasmid transformed into E. coli will contain antibiotic resistance. Plating out
Aldrich transformed cells onto antibiotic inoculated medium will only grow cells that have
been successfully modified.
E. coli from ATCC $50 Chemically competent E. coli strain
DNA Extraction kits $400 Mo-Bio Mo-Bio soil DNA extraction kit 50 uses (2 kits)
Genetic facilities use $1,300 University This encompasses all PCR, gel electrophoresis, and imaging facilities use. This also
of Guelph includes reagents such as Taq polymerase, and nucleotide stock solutions (as master
mix), and fluorescent indicator (SyberSafe).
Restriction enzymes $200 Invitrogen Eco R1 and Hind III restriction enzymes to cut DNA at key points
DNA ligase enzyme $200 Invitrogen Ligase enzyme, in order to fuse restriction digested DNA fragments to restriction
digested plasmid DNA.
PCR primers $200 Lab Services Primers for arsenic methylation gene conserved regions (from BLAST database). This
will likely take some refinement, but will include restriction sites to flank the gene of
interest. These restriction sites are specific for restriction enzymes, which cleave the
dsDNA in a staggered manner, and allow it to be reattached using a ligase enzyme.
DNA sequencing $150 Lab services DNA sequencing of PCR products after electrophoretic separation
Nanodrop facility $150 Lab services Low quantity DNA quantification technique
Methylation reaction reagents $250 Sigma Necessary substrates for methylation reaction (after plasmid is cloned into E. coli and
Aldrich protein is expressed and functional).
TLC separation reagents $100 Sigma To separate methylated arsenic species using the method of Styblo et al (1995) J.
Aldrich Chrom. B 668: 21-29, using thin layer chromatography for quick confirmation
Plasmid kit $700 Invitrogen Necessary for transfer of target gene from DNA extractions and PCR amplification to
E. coli
DNA ladder for gels $200 Invitrogen DNA ladder to identify molecular weight of DNA products on an electrophoresis gel
Travel to Fleming $330 Car rental and per diem for 3 trips

Fed Dev ARCI funding is $50,000 of cash with $25,000 of in-kind services donated by Nature Works for this funding. Activities must be completed by March 31, 2012.
The deadline to start was originally March 31, 2011 but has been extended to June. But we want to get started asap.
Nature Works and Kinross are also partners under my NSERC IE grant and are eligible for support under it.
Problem is that Fed Dev has been extremely clear that we cannot use funding from other projects to extend or enhance what their funding is doing in any way.
If we do use funding to top up the Fed Dev project and they catch us they'll remove that amount from the Fed Dev award.
We have to have a discrete $50,000 project. After it is finished we can do what we like for a different project under a different name.
Other issues - I cannot send any funding from my NSERC grant to universities. So I would have to employ any labour directly from Fleming.
Fleming is unionized and our pay rates are higher so this is a disincentive if the budget is tight.
What we need to do is find a 3-6 month discrete project that costs $50,000 (with in-kind services provided by Nature Works so we can claim the funding).
