         Novel Feed Ingredients for Trout
             and Hybrid Striped Bass

    Description

            The determination of nutrient digestibility’s in specific ingredients
    and diets for fish has been an area of active research for decades. The
    Apparent Digestibility Coefficients (ADC), the percentage of nutrients in an
    ingredient that are available to the fish, is information needed by
    researchers, producers, and feed mills to accurately formulate feeds and
    thus meet the needs of the animal without excess. ADC’s are also
    necessary for determining the nutritional and economic value of alternative
    ingredients. Data developed from many different laboratories have been
    compiled in publications such as the NRC (1993), and often show extreme
    variability. This is not unexpected since there are many factors that can
    affect the ADC of an ingredient, including basal diet formulation, method of
    feed manufacturing (cooking versus cold formation), fecal collection
    method, etc. Different laboratories often use a mixture of methods specific
    to that laboratory.


           A project was initiated in 2005 to determine ADC’s for
    macronutrients, amino acids and minerals for traditional and novel
    ingredients in rainbow trout and hybrid striped bass, using standardized
    conditions as much as possible. On this spread sheet, the content of            H
    macro-nutrients amino acids and minerals of each ingredient is provided
    on worksheet labeled Nutrient Composition. The ADC’s for each
    ingredient for specific nutrients are found on the worksheets labeled “ADC,
    Trout” or alternatively, “ADC, HSB” for hybrid striped bass. The anti-
    nutrient composition of selected ingredients is found on worksheet labeled
    Anti-nutrients. The database will be updated as additional ingredients are
    evaluated. The intention of this project was not to be all inclusive of every
ingredients in rainbow trout and hybrid striped bass, using standardized

Procedures
               The nutritional value of each ingredient was evaluated by
determining the apparent digestibility of nutrients and energy and amino
acid availability in compounded, extruded diets. Nutrient and energy
availability were determined using the methods described by Cho et al.
(1982) and Forster (1999) to estimate apparent digestibility coefficients
(ADCs). All diets were double labeled using chromic oxide and yttrium
oxide as the inert markers. A complete reference diet meeting or
exceeding all known nutritional requirements for rainbow trout and hybrid
striped bass was blended with the test ingredients in a 70:30 ratio (dry-
weight basis) to form test diets (Tables 1-3). The reference diet has
changed during the project to improve accuracy of phosphorus
determination or improve feed intake with hybrid striped bass. These
reference diets have been used successfully in several digestibility and
growth trials with rainbow trout (Barrows et al., 2008; Gaylord et al., 2008;
Gaylord et al., 2009; Barrows et al., 2009; Gaylord et al., 2010).

              Apparent digestibility coefficients of each nutrient in the test
diet and ingredients will be calculated according to the following equations
(Kleiber 1961, Forster 1999):

where,
ADCNingredient = apparent digestibility coefficient of the nutrient in the test
                  ingredient.
ADCNt = apparent digestibility coefficients of the nutrient in the test diets.
ADCNr = apparent digestibility coefficients of the nutrient in the reference diet.
a = (1-p) x nutrient content of the reference diet.
b = p x nutrient content of the test ingredient.
p = proportion of test ingredient in the test diet.

       All diets were manufactured by cooking extrusion (DNDL-44, Buhler
AG, Uzwil, Switzerland). General operating conditions included
approximately 18-s exposure to an average of 127oC in the six extruder
barrel sections. The exception is experiment 6 in which diets were
produced using cold-forming methods as denoted on the database. For
the cooked, extruded diets, pressure at the die head varied from 260 to
450 psi, depending on test ingredient. Pellets of 3-4 mm were produced
and dried in a pulse-bed drier (Buhler AG, Uzwil, Switzerland) to less than
10% moisture, followed by a 20 minute cooling period with forced air at
ambient temperature. Diets were top-coated with fish oil using a vacuum
coater (A.J. Mixing, Ontario, Canada) at 24 mm mercury pressure.


       The experimental diets were fed to trout held in 140 or 500-L tanks
and hybrid striped bass held in 630-L tanks. Fish sized varied across
experiments and covered a range from 250 to 600 g for rainbow trout and
75 to 150 g for hybrid striped bass. Stocking density also varied
accordingly with 10-15 rainbow trout per tank or 30-50 hybrid striped bass.

         The experimental diets were fed to trout held in 140 or 500-L tanks
  and hybrid striped bass held in 630-L tanks. Fish sized varied across
  experiments and covered a range from 250 to 600 g for rainbow trout and
  75 to 150 g for hybrid striped bass. Stocking density also varied
  accordingly with 10-15 rainbow trout per tank or 30-50 hybrid striped bass.
  Water temperature was maintained at 15 ºC while lighting was maintained
  on a 13:11 h diurnal cycle for rainbow trout. Water temperature for hybrid
  striped bass ranged from 19-23 ºC and lighting was maintained on a 12:12
  diurnal cycle. Each diet was randomly assigned to three different tanks of
  fish and fed to satiation by hand twice daily. Fecal samples were obtained
  by manual stripping, 16-18 h post-feeding for trout or 6-8 h post-feeding
  for hybrid striped bass. Manual stripping of rainbow trout in each tank was
  accomplished by netting and anesthetizing the fish, followed by gently
  drying and then applying pressure to the lower abdominal region to first
  express urine to a waste container and then fecal matter into a plastic
  weighing pan. Hybrid striped bass were similarly stripped without the use
  of anesthetics. Fecal samples for each tank were dried either by heating
  at 50ºC overnight in the early trials or by freeze-drying in the later trials.
  Samples were stored at -20 0C until chemical analyses were performed.




  Adjustment of ADC values
         A variety of factors can result in negative ADC values or values
  above 100%. This most often occurs when the nutrient is present in very
  low levels in the test ingredient (e.g., fat in solvent extracted soybean
  meal) or has a true digestibility near to either threshold. In each of these
  cases the values were rounded down to 100% or up to zero for feed
  formulation purposes.
