The titration of an amino acid



These data represent the pH titration curve of an amino acid. A plot of pH against

volume of base added produces the classic S-shaped curve expected. The values of

pK1, pK2 and the Ionisation Potential (pI) can be obtained from the graph. A more

accuarate esitmate of pI can be achieved using a derivative plot -- that is a plot of

rate of change of pH against volume (see below).

Using a derivative plot



Here the idea is to graph the rate of change (i.e. first derivative) of pH against volume of

base added. We start with the same data as before:

You now need to plot the derivative. If you have followed the instructions above you will

have three columns of data. But you only want to plot the first (volume) and last (rate of

change of pH). To plot these data:



(1) Select the data in the first column in the usual way, then, while it is still selected press

and hold down the Ctrl key.

(2) Now select the data in the second column.

(3) Call the chart wizard and edit your graph.
