Bead- Based Automated Phage Selection

Abstract:
Phage Selection is a powerful tool for identifying competitive inhibitors for small molecule high
throughput screening assays. Using a bead-based system it should be possible to automate a 10
round selection in 24 hours to routinely select a “global” maximum. A photocleavable linker
attaches the target protein through lysine residues that enables unbiased elution with respect to
phage binding properties. The resulting peptide ligands with an appropriate reporter group, such
as fluorescein, can be used to in order to screen small molecules against the targeted protein in a
high throughput manner.
