X-Ray Protocol
Use plastic bottles. Glass bottles cause scattering of X-rays
For patches in the eye use the 1200R setting for embryos/larvae that are 12-36 hours old.
Start cross at 11 PM.
Let the flies lay for 24 hours.
Transfer the flies out at 11PM.
X ray the flies at 11 in the morning.
Warm up the machine for 15 minutes. Turn on the machine (use power breaker on wall
and use the key), turn the timer off, let the machine stay on with the X-rays on for 15
minutes (call ahead to have someone in the Zip lab do this for you)
Turn the X-ray key to safe.
Center the bottle on the clamp inside the X-ray machine. Look at the X on the ceiling.
Make the surface of the food 27cm (10 3/4 “) below the ceiling (use the ruler on the
keychain).
Mutagenize at 2’ 28”...100kV...20mA ---> 1000R
2’ 58”...100kV...20mA ---> 1200R (use this setting for patches)
Make sure the Max mA is off. Make sure the focus is on the wide square.
(use the pre-indicator button to get 100kV)
Make sure the door is slammed shut.
Turn the key all the way to the X-ray.
Hit the start button with the timer set for appropriate time.
