Tissue culture supernatant diluted in 5% horse serum in PBS containing 12mM sodium azide. Volume as indicated on vial label. Immunohistochemistry: Typical working dilution: NEAT. 15 minutes primary antibody incubation at 25oC when used in conjunction with the VECTASTAIN® Universal Quick Kit (R.T.U.), code PK-7800. Recommendations on use will differ if other detection systems are used eg Standard ABC technique. Not recommended for use on Ventana automated staining systems (Ventana Medical Systems Inc., USA). Positive controls Adrenal gland. The medulla shows intense staining, contrasting with the cortex which should be entirely negative. Normal bowel. The enterochromaffin cells are located in small numbers on the outer aspect of glands and show staining for chromogranin contrasting with surrounding epithelial cells and smooth muscle which should be negative. Tissue from stomach, small or large bowel can be used. Normal thyroid. The C (calcitonin) cells between the follicles, arranged in small groups, are positive for chromogranin. Staining pattern Storage and stability Fine granular cytoplasmic staining. Store ready to use prediluted liquid antibody at 4oC. Return to 4oC immediately after use. Chromogranin A is a 68kD acidic protein. In normal tissues, chromogranin A is widely expressed in neuronal tissues and in secretory granules of human endocrine cells eg parathyroid gland, adrenal medulla, anterior pituitary gland, islet cells of the pancreas and C cells of the thyroid. RTU-CHROM is useful for the identification of neuroendocrine tumours eg pituitary adenomas, islet cell tumours, phaeochromocytomas, medullary thyroid carcinomas, Merkel cell tumours and carcinoids.
